Gardner-Webb (9-19) took a 55-54 lead on Stefon Johnson's free throw with 1:58 left, but Murphy hit back-to-back jumpers and then added two free throws to give Tennessee Tech a 60-55 lead with 2 seconds to play.
Luke Engelken hit a 3-pointer at the buzzer for the final margin.
The Golden Eagles won despite shooting 39.5 percent from the field (17 of 43) and committing 17 turnovers.
Gardner-Webb led 45-38 on Santoine Butler's dunk with 8:34 left, but Tennessee Tech scored the next eight points for a 46-45 lead with 6:38 to play.
Laron Buggs scored 19 and Jon Moore added 15 points for Gardner-Webb.
