PSV Eindhoven failed to reclaim top spot in the Dutch league after losing 3-1 at Feyenoord on Sunday, dropping to third following Ajax's 4-1 win over Excelsior Rotterdam.
FC Twente went top after beating ADO The Hague 2-1 on Friday, and the defending champions will be bidding to stop Ajax winning the title for a record 30th time.
With two rounds remaining, Twente is one point in front of Ajax and three ahead of PSV.
Twente visits Ajax on the last day of the season in two weeks' time. The sides also meet in the Dutch Cup final on May 8 at Rotterdam Kuip stadium.
Lorenzo Ebecilio scored from a rebound to give Ajax the lead in the 13th minute, but Daan Bovenberg equalized in the 54th after combining with Ryan Kool.
Ajax regained the lead when Gregory van der Wiel set up Christian Eriksen in the 72nd. Teammate Ebecilio added another four minutes later, and Siem de Jong wrapped up the win in injury time.
PSV captain Orlando Engelaar was sent off in the 54th for a foul on Luc Castaignos, ruling him out of the home match against Vitesse Arhem on Sunday. Four other PSV players will miss the match after each picked up their fifth yellow card of the season.
Georginio Wijnaldum put Feyenoord ahead in the 27th minute. Ola Toivonen equalized from a long-range shot in the 60th, but Wijnaldum scored again with a header from Diego Biseswar's cross three minutes later.

Also Sunday, Michihiro Yasuda, Dries Mertens and Kevin Strootman scored in FC Utrecht's 4-2 win over Vitesse Arnhem. Guram Kashia replied for Vitesse in the 54th. Frank Demouge and Adnane Tighadouini were also on target for either side.
