Shoni Schimmel had 14 points and Asia Taylor added 12 points and 10 rebounds for Louisville.
Five different players scored early - including eight points from Martin - as DePaul burst to a 17-2 lead through the opening 6:26.
DePaul, limited by injuries to a seven-player rotation, opened its biggest lead at 77-47 on Deanna Ortiz' 3 pointer with 5:40 to play.
DePaul improved to 10-1 at home, the only loss coming against No. 3 Connecticut.
